Abstract:In this research, dual-color fluorescence in situ hybridization ( FISH) on Jimai serial wheat was performed using Oligo-pTa535-1 and Oligo-pSc119.2-1 as probes. As a result, the FISH signals of Oligo-pTa535-1 mainly distributed on chromosome A and D, while the FISH signals of Oligo-pSc119 .2-1 mainly distributed on chromosome B.The standard FISH patterns of Jimai varieties ( lines) were somehow different from those of Chinese Spring, and the different FISH sites were mainly distributed on chromosome 4A, 6B, 7B and 1D, mostly were signals of Oligo-pSc119.2-1.The signal of Oligo-pTa535-1 on cen-tromeric region of chromosome 1B of Jinan17, and the signals of Oligo-pSc119.2-1 on terminal region of chromosome 1D short arm and chromosome 2A long arm of Jimai20 could be regarded as their cytology tags. The establish of standard FISH patterns of Jimai serial wheat laid good cytogenetic foundation for wheat im-provement through chromosome engineering. The similarity of FISH signal variation positions of Jimai serial wheat indicated a narrow genetic bases of them. Therefore, Jimai serial wheat should cross with parents pos-sessing relatively distant genetic relationship in wheat breeding in the future.
