Genetic Diversity Analysis and DNA Fingerprint Construction of Peanut Genotypes Resistant to Aspergillus flavus

Abstract:To study the DNA polymorphism and genetic relationship of peanut germplasms with resistance to Aspergillus flavus and explore excellent genotypes, the genomic DNA isolated from 48 peanut lines related to Aspergillus flavus invasion and aflatoxin production were amplified using 46 pairs of polymorphic SSR markers. The results showed that the average alleles revealed by each pair primers were 5.15 and the average PIC value was 0.604.The 48 lines were divided into eight groups by UPGMA cluster analysis and resistant germplasms were distributed into five groups.Among which, low aflatoxin producing line 91048, invasion-resistant line AR-3, PI337409F and GFA -2, and Thielariopsis basicola -resistant line PERRY had obvious genetic differences with other germplasms, which could be used as ideal breeding materials.The fingerprint profiles based on“primer +0 ,1 character string” of 48 lines were constructed using 35 alleles of 5 SSR markers.The screened excellent germplasms could be used as parents in the breeding of peanut genotypes resistant to Asper-gillus flavus and broad its genetic basis.
