In silico Cloning and Bioinformatics Analysis of Voltage -Dependent Anion Channel Gene NtVDAC1.1 from Nicotiana tabacum

Abstract:In order to investigate the function of voltage -dependent anion channel (VDAC)gene in to-bacco root secreting organic acid and provide basis for its research and application,a full -length cDNA se-quence of VDAC gene,termed as NtVDAC1.1,was obtained using AtVDAC1 (NM_1 1 0994)as search probe. Bioinformatics analyses showed that the full length of NtVDAC1.1 was 1 01 8 bp,which contained a complete open reading frame and encoded 276 amino acid residues.Homology and phylogenetic relationship analyses suggested that NtVDAC1 .1 shared high similarity with VDAC of other plant species,and the identities with three porins from Solanaceae potato were 0.768 ~0.902.NtVDAC1 .1 belonged to porin,which took part in anion transport,regulation of anion transport and transmembrane transport.The prediction of secondary and tertiary structure indicated that NtVDAC1 .1 had α-helixs and β-sheets.Amino acid site analysis suggested that NtVDAC1 .1 possessed a number of types of modification sites.These phosphorylation sites indicated that the protein might take part in plant signal transduction and regulation of anion channel activity.
