Cloning and Bioinformatics Analysis of TaXTH from Wheat -Thinopyrum Alien Addition Line

Abstract:In this study,a novel gene fragment up -regulated in response to 20% PEG -6000 was i-dentified from the cDNA of wheat -Thinopyrum alien addition line SN6306 by GeneFishing technology,which was named 7154 and had the length of 586 bp.Through the sequence alignment,it was predicted as a part of xyloglucan endotransglucosylase /hydrolase gene (XTH).The full cDNA and DNA sequences of the putative TaXTH were obtained by using cloning and RT -PCR.The sequence analysis of the nucleic acid and the pre-dicted protein showed that the full DNA sequence of TaXTH was 2 224 bp,and it contained 3 exons and 2 in-trons;its open reading frame (ORF)was 936 bp,and encoded 311 amino acids.The BlastP analysis showed that TaXTH belonged to GH16 protein family and had 79% identities with the homologous protein from Brachypodium distachyon.Based on the results of multiple sequence alignment and polygenetic tree analysis, it was inferred that TaXTH belongs to XTH and had the typical conservative DEIDFEFLG motif.
