Optimization of Sporulation Fermentation Process of Bacillus subtillus NS178 by Response Surface Methodology

Abstract:Three factors mainly influencing the colony number of endophytic Bacillus subtillus NS178 in fermentation liquid were screened out by single-factor experiments, which were glucose, ( NH4 ) 2 SO4 and K2 HPO4 .The Box-Behnken Design was applied to optimize the fermentation medium for NS178, and the op-timum one was obtained as 2.29%glucose, 0.3%(NH4)2SO4 and 0.23%K2HPO4 after the analysis of re-sponse surface methodology.The culture conditions were also optimized, and the optimal conditions were tem-perature as 33℃, pH value as 7.6 and fluid volume as 50 mL.Under the optimal fermentation conditions, the yield of NS178 was 34.50 ×108 cfu/mL;the spore rate could reach 75.8%after cultured for 72 hours;and the diameter of inhibition zone increased by 67%compared to that before fermentation.
