Cloning and Expression Analysis of AhHSP70 and AhHSF Genes in Arachis hypogaea L.

Abstract:Heat shock proteins ( HSPs) and heat shock factors ( HSFs) play important roles in the plant response to high temperature stress.In this study, the cDNA fragments of HSP70 and HSF genes were ob-tained from peanut transcriptome sequences, and through sequence alignment, their genomic sequences were found in the whole genome sequence of peanut.Based on the predicted coding sequences, the primers were designed, and then the full-length ORFs were amplified using the peanut leaf cDNA as template.The se-quence analysis indicated that the ORF of AhHSP70 was 1 962 bp encoding a protein of 653 amino acids;its molecular weight was 71.45 kD and the theoretical pI was 4.93.The ORF of AhHSF was 1 212 bp encoding a protein of 403 amino acids;and its molecular weight was 46.03 kD and the theoretical pI was 4.85.AhH-SP70 and AhHSF were both soluble proteins, and had no signal peptide;there were a large number of random coils in their secondary structure.The phylogenetic analysis revealed that AhHSP70 shared higher homology with Glycine max HSP70, but lower with Solanum lycopersicum HSP70;AhHSF shared higher homology with Phaseolus acutifolius HSF but lower with Medicago truncatula HSF.The quantitative real-time PCR was used to analyze the expression of AhHSP70 and AhHSF genes after heat treatments at 42℃.The results showed that transcripts of these two genes were significantly induced by high temperature.The expression of AhHSP70 was significantly up-regulated after 3 hours of treatment,and the expression levels at 24 h and 48 h were 50 and 135 times of the control.The expression of AhHSF was also significantly up-regulated after 3 hours of high temperature treatment, and reached the maximum expression level at 6 h, then declined.This study prelimi-narily validated that the two genes played important roles in peanut response to high temperature stress.
