Study on Regeneration System of Lycopersicon esculentum Mill

Abstract:The effects of different culture medium , hormone concentration and ratio on callus induction , adventitious bud differentiation and root induction of Lycopersicon esculentum Mill.were studied by culturing cotyledon and hypocotyl in vitro.Vc was added in culture medium to control browning phenomenon .The re-sults showed that the optimum medium and hormone ratio for callus induction was MS+(0.2~0.4) mg/L IAA+2 .0 mg/L 6 -BA; the optimum medium and hormone ratio for adventitious bud differentiation was MS+0.1 mg/L IAA+(1.0~3.0) mg/L 6-BA;the optimum medium and hormone ratio for root induction was 1/2MS+0.1 mg/L NAA.Cotyledon was the best explant , which showed higher differentiation speed and rate, and better bud growth condition than hypocotyl .The browning phenomenon could be controlled through adding 10 mg/L Vc in callus induction medium .
