Comparative Study on Extraction Methods of Total RNA from Leaves of Acer Truncatum ‘Luhong No.1’

Abstract：A large amount of polysaccharides and polyphenols in Acer truncatum‘Luhong No.1’leaves have seriously affected the yield and quality of total RNA extraction .In order to obtain the appropriate method of total RNA extraction,3 methods,Kit method,TRIZOL and modified CTAB,were chosen to extract them from Acer truncatum‘Luhong No.1’autumn leaves.The results showed that the total RNA extracted by Kit method had the general yield,the ratios of OD260/OD280 and OD260/OD230 were both above 1.8,and had the pollution of DNA.That extracted by TRIZOL method had the lowest yield ,the ratios of OD260/OD280 and OD260/OD230 were both about 1.5.That extracted by modified CTAB method showed clear electrophoresis pattern and highest yield,and the ratios of OD260/OD280 and OD260/OD230 were both about 2.0.The results demonstrated that the yield and purity of the RNA obtained by the modified CTAB method could meet the demands of molecular biol -ogy experiment .The modified CTAB method was the appropriate method for total RNA extraction from Acer truncatum‘Luhong No.1’ leaves.
