Cloning of SnRK2.2 Gene from Wheat, Construction of Expression Vector and Transformation

Abstract:Sucrose non-fermenting 1-related protein kinase 2 ( SnRK2 ) gene family members partici-pate in the process of protein phosphorylation and play essential roles in response to abiotic stress in plants .In this study , a new member of SnRK2 in wheat named TaSnRK2 .2 was cloned by RT-PCR method based on the nucleotide sequence of SAPK2 gene in rice .The new sequence was submitted to GenBank and the acces-sion number was KJ850253 .TaSnRK2.2 gene was 4 118 bp containing nine exons and eight introns with an open reading frame of 1 026 bp which encoded 341 amino acids.The sequences of SnRK2.2 gene showed highly conservative in gramineous plants , and TaSnRK2 .2 had closer relationship with SAPK2 from rice and SnRK2.2 from maize.The molecular weight of TaSnRK2.2 protein was 38.64 kD and the theoretical pI was 5 .45 .It had typical serine/threonine kinase conserved domain and several phosphorylation sites .The over-expression vector of TaSnRK2.2 gene was constructed and transformed into Arabidopsis thaliana, and the transgenic plants were obtained .Stable expression of TaSnRK2 .2 gene was detected in transgenic plants .
