Optimization of Culture Conditions for Aseptic Sweet Pepper Seedling

Abstract：Using Qiemen sweet pepper as materials , the disinfection effects of different disinfectant a-gents, disinfection times and contents on seeds before culture were compared , and the effects of medium for-mula, pH value and growth time on growth of aseptic seedling were investigated .The results showed that the optimum disinfection method and culture conditions for aseptic sweet pepper seedling were soaking seeds in a -septic water of 55℃for 15 minutes, then staying at room temperature for 4~5 hours, disinfecting with 10%NaClO for 30 minutes, washing 3 times with aseptic water and then culturing in medium of 1/2MS+15 g/L sugar+7.5 g/L agar with pH value of 5.8 under (25 ±1)℃.When the cotyledon sprouted for 6~10 days, the seedlings could be used for genetic transformation .
