Establishment of Soybean Cotyledon Node Transgenic Receptor System

Abstract：Using three soybean varieties for materials,the effects of seed disinfection method,different hormone concentrations at bud induction and rooting stages on the regeneration of soybean cotyledon node were studied to establish an efficient regeneration system for soybean genetic transformation.The results showed that the best seed disinfection method was disinfecting with 70%alcohol for 30 seconds,then HgCl2 for 7 minutes, and finally washing 3~5 times with sterile water.The optimum bud induction medium for Jinong 28 was MS+1.5 mg/L 6-BA +0.5 mg/L IBA;for Jinong 27 and Jiunong 21,MS+2.0 mg/L 6-BA+0.8 mg/L IBA was the best.During the rooting stage,the best rooting mediums for three soybean varieties were all 1/2MS +2.0 mg/L IBA.
