Isolated Microspore Culture and Plant Regeneration of Chinese Cabbage

Abstract:The factors affecting isolated microspore culture and plant regeneration were studied with four Chinese cabbage varieties of‘Fushanbaotou’.The results showed that there were significant differences in embryogenesis ability of microspore and seedling rate among different genotypes.The embryo yield per bud ranged from 1.28 to 38.25.The varieties with higher embryogenesis ability had higher seedling rate,while those with lower embryogenesis ability had lower seedling rate.The addition of 0.1 ~0.3 g/L active charcoal could increase the induction rate of microspore-derived embryos significantly.Transferring mature cotyledon-ary embryos to regeneration medium in time was essential to increase plantlet regeneration rate.The solid medi-um with agar concentration from 1 .0% to 2.0% could increase seedling rate obviously.
