Study on Factors Influencing Electro-Transformation Efficiency of Saccharomy cescerevisiae

Abstract：Using S.cerevisiae EGY48 as material, the effects of different incubation times (12, 16, 20, 24 and 30 h), cell conditions (OD600), electric voltages (600, 800, 1 000, 1 200 and 1 400 V) and plas-mid DNA dosages (1, 2, 5, 10, 15 and 20μg) on transformation efficiency were studied .The results showed that when the incubation time was 20 h, OD600 value was 1.4, electric voltage was 1 000 V and plasmid dos-age was 5μg, the transformation efficiency was the highest , which laid a foundation for further screening of ef-ficient library .
