Study on Quality Control and Identification Techniques of Radix Codonopsis Based on Electrospray Ionization Ion Mobility Spectrometry

Abstract:Objective:Aiming at Radix Codonopsis,a representative of Chinese herbal medicines and also of a complex sample system,to develop an efficient quality control and identification technique based on electro spray ionization ion mobility spectrometry (ESI-IMS). Methods:Methanol was used to extract the components of Radix Codonopsis,and the two-dimension ion mobility spectra were obtained by using ESI-IMS. The con-trol charts for quality fluctuation analysis of Radix Codonopsis were constructed based on the previous ES-I-IMS spectra. The two-dimensional correlation ESI-IMS spectra as obtained by changing the ion gate pulse width. Results:The typical two-dimension ion mobility spectral peaks of Radix Codonopsis were at 9.5 ms and 10 ms. In the average control charts for quality fluctuation analysis of Radix Codonopsis,the center line was at 0.7811,with the upper control limit 1.284,and the lower control limit 0.2783,within the scope of the normal distribution. The two-dimensional correlation ESI-IMS spectra showed the feature information of Radix Codonopsis in the three-dimensional space. Conclusion:Through the three different aspects of the evaluation analytical method,a new system of quality control and identification technique of Radix Codonopsis has been constructed and can be applied for the identification and quality control of Radix Codonopsis.
