Study of Tyrosine Phosphorylation and Protein Expression of Insulin Receptor Sub-strate-1 in Ovarian Luteinizing Granulose Cells from the Patients with Polycystic Ovary Syndrome of Phlegm-dampness Syndrome

Abstract:Objective:To explore molecular mechanisms of insulin resistance and ovary reproductive dysfuction by determining tyrosine phosphorylation and protein expression of insulin receptor substrate(IRS)-1 in ovarian luteinizing granulose cells from the patients with polycystic ovarian syndrome (PCOS) of phlegm-dampness syndrome. Methods:Ovarian luteinizing granulose cells were obtained in the process of in vitro fertiliza-tion-embryo transplantation from the 77 patients. The patients with PCOS of phlegm-dampness (n=21),PCOS with non phlegm-dampness(n=26) and subjects with normal ovulation(as control,n=30). Serum erythropoietin FINS was measured by chemical light method. FPG was measured by oxidase assay.Insulin resistance index was calculated using homeostasis model assessment(HOMA-IR). The tyrosine phosphorylation and protein ex-pressions of IRS-1 in ovarian luteinizing granulose cells were analyzed by Western blot. Results:1.Comparison of related parameters of insulin resistance in patients of Each group:Compared with control group,two groups FPG,FINS,HOMA-IR of PCOS were increased(P<0.01);Compared with non-phlegm-dampness group of PCOS, FINS and HOMA-IR in PCOS phlegm-dampness group were increased(P<0.01). 2.Comparison of the degree tyrosine phosphorylation and protein expression of granular cell IRS-1 in each group:the protein expression of IRS-1 in two groups of PCOS patients were significantly higher than those in the control group(P<0.01), and PCOS phlegm-dampness group was significantly lower than non-phlegm-dampness group(P<0.05). IRS-1 tyrosine in two groups of PCOS were significantly lower than those in control group(P<0.01),and PCOS phlegm-dampness group was significantly lower than non-phlegm-dampness group(P<0.01). Conclusions:Local area of ovarian in patients of phlegm-dampness PCOS group had insulin resistance and ovary reproductive dysfunction. The possible reason may be related with anomalies of protein expression of IRS-1 and tyrosine phosphorylation. This study provided objective basis for the diagnosis of PCOS phlegm-dampness syndrome.
