Effects of Bushen Huoxue Formula on Proliferation Apoptosis of Chondrocyte and MMP-13 Level

Abstract：Objective:To determine the effect of Bushen Huoxue formula(BHF) on chondrocyte apoptosis and MMP-13 level in rat chondrocyte in vitro. Methods:Cartilage from sprague dawley rats were used for cell culture in vitro. Toluidine blue stain and Ⅱ immunohistochemistry stain were used for identification. After cell synchronization,cells were divided into Chinese herbal medicine group,western medicine group,blank control group. Flow cytometry was used for cell apoptosis and proliferation ,and MMP-13 level were deter-mined. Results:Cell culture and identification of chondrocyte was successful. BHF was confirmed effectively reduced the apoptosis of chondrocytes as well as increased the proportion of G2 and S stage cells with ex-tremely significant difference compared to the blank control group (P<0.01). Meanwhile,BHF could reduce the MMP-13 level with significant difference compared to blank control group(P<0.05). Conclusion:BHF has the effects of reducing the apoptosis rate and MMP-13 level of chondrocytes.
