The role and regulatory mechanism of oxidative stress system in drug hepatotoxicity

Abstract:Drug-induced liver injury(DILI)is a core issue that leads to the interruption of drug development and clinical treatment risks,and its pathological mechanism is closely related to the imbalance of the oxidative stress system.This article first clarifies the generation and toxicity of reactive oxygen species(ROS)and the hierarchical regulatory mechanism of the antioxidant defense system.Further,it deeply explores the correlation between oxidative stress and DILI.Finally,it summarizes the regulatory mechanisms and intervention strategies for targeting the oxidative stress system to alleviate DILI.This review integrates the multi-dimensional correlations between the oxidative stress system and the pathological process of DILI,providing theoretical support for the prevention and treatment of DILI.
