Research status and progress on DNA vaccines for chronic hepatitis B

Abstract:Chronic hepatitis B(CHB)is a chronic infectious disease caused by the hepatitis B virus(HBV),with a high global prevalence.Current antiviral therapies primarily rely on traditional intramuscular injection drugs,which still face limitations such as incomplete viral clearance,drug side effects,drug resistance,and the need for long-term or lifelong medication.As an emerging therapeutic strategy,DNA vaccines have garnered significant attention due to their potential to simultaneously induce humoral and cellular immune responses and their high stability.Preclinical studies in animal models have demonstrated promising results.By combining adjuvants and electroporation techniques,the uptake efficiency of plasmid DNA can be enhanced,significantly improving therapeutic efficacy.Preliminary results from related clinical trials have also been encouraging.This review summarized the current research progress of DNA vaccines in CHB treatment,including their mechanisms of action,preclinical studies in animal models,clinical trials,distinctions from RNA vaccines,and future development directions.
