Efficacy and safety of Donepezil Hydrochloride Tablets in the treatment of dementia:a network Meta-analysis

Abstract：Objective To assess the effectiveness and safety of both domestic and international donepezil in treating dementia via a network Meta-analysis,offering a more dependable guide for clinical medication choices in managing demen-tia.Methods An extensive search was performed across PubMed,Embase,Cochrane Library,CNKI,WanFang Data,and CBM databases to gather randomized controlled trials(RCTs)that evaluated domestic versus imported donepezil for dementia treatment,covering the period from January 1996 to January 2024.Two investigators separately reviewed the litera-ture,collected data,and evaluated the potential for bias in the studies considered.A network meta-analysis was conducted u-tilizing Stata 17.0 software.Results 34 studies with a combined total of 3 432 participants were included.The outcomes of the network meta-analysis indicated that domestic donepezil(Surface Under the Cumulative Ranking curve,SUCRA=0.983)was more effective in treating dementia than imported donepezil(SUCRA=0.748).However,imported donepezil(SUCRA=0.855)showed better MMSE scores than domestic donepezil(SUCRA=0.640).The incidence of adverse reactions was slightly lower with imported donepezil(SUCRA=0.537)compared to domestic donepezil(SUCRA=0.499).Conclusion Current evidence suggests that the overall efficacy and safety of imported and domestic donepezil are similar.Nonetheless,because the quantity and caliber of the studies included are restricted,additional large-scale,high-quality re-search is necessary to confirm these results.
