Research progress of dsRNA in anti-tumor immunity

Abstract:Double-stranded RNA(dsRNA)is a nucleic acid molecule with significant biological functions,widely present in viral genomes and host immune response processes.In recent studies,dsRNA has been identified as a potent acti-vator of immune responses,capable of triggering a series of signal transduction pathways by engaging pattern recognition re-ceptors(PRRs).This activation leads to the induction of type Ⅰ interferons(IFNs)and pro-inflammatory cytokines,ulti-mately eliciting robust immune responses.Consequently,dsRNA has emerged as a critical focus in cancer immunotherapy re-search.This review systematically summarized the latest advancements in the role of dsRNA in antitumor immunity,with a particular emphasis on its mechanisms of action,including the activation of dsRNA-sensing pathways,viral mimicry,and re-modeling of the tumor microenvironment.Furthermore,the review explored the potential applications of dsRNA in cutting-edge therapeutic strategies,such as cancer vaccine development,combination therapy with immune checkpoint inhibitors,and RNA interference(RNAi)-based therapies.By synthesizing a comprehensive analysis of these mechanisms and applica-tions,this review aims to offer clearer research directions and insights for the further utilization of dsRNA in cancer immuno-therapy.
