Study on the protective effect of Tibetan medicine Gentianopsis paludosa Ma on NCM460 cells

Abstract:Objective To investigate the protective effect of Gentianopsis paludosa Ma extracts(GPE)on human normal colon epithelial cells(NCM460).Methods Preparing GPE and establishing an inflammation injury model using LPS-induced NCM460 cells.Subsequently,GPE(50,100,200 μg·mL-1)was administered for 24 hours,and cell viability was as-sessed using the CCK-8 kit.Using Western blotting and qRT-PCR techniques,the protein and gene expression of zonula oc-cludens protein-1(ZO-1),Claudin-3,and Occludin in NCM460 cells were analyzed,and flow cytometry was used to label apoptotic cells and evaluate their apoptosis status.Results After 24 hours of stimulation with 10 μg·mL-1 LPS,the survival rate of NCM460 cells dropped to below 80%,indicating that the inflammation injury model was successfully established.The intervention using GPE significantly improved cell survival rates and further increased the protein and gene expression levels of tight junction proteins ZO-1,Claudin-3,and Occludin in the cells.Moreover,GPE also significantly inhibited the apoptosis rate of cells,with the most notable effect at a concentration of 200 μg·mL-1.Conclusion GPE exerts its protective effect by effectively inhibiting the degradation process of tight junction proteins in NCM460 cells and alleviating abnormal apoptosis.
