Study on the inhibitory effect of vitexin and isovitexin on pancreatic lipase

Abstract：Objective To investigate the inhibitory effect of vitexin and isovitexin.Methods In vitro experiments was applied to simulate fat digestion,and the inhibition types of them on pancreatic lipase were fitted by enzyme reaction kinet-ics,molecular docking technology was used to explore the interaction between the two and pancreatic lipase.Results The half inhibitory concentrations of vitexin and isovitexin were 268 μg·mL-1 and 221 μg·mL-1,respectively.The molecular doc-king results showed that the comprehensive scores of their binding to pancreatic lipase were 5.173 0 and 6.125 1,respective-ly.The binding forces included van der Waals forces,hydrogen bonds,C-H bonds,π-bond interactions,and hydrophobic forces.In addition,when the mass concentration ratio of the two was 1∶0.7,the drug combination index(CI value)for pancre-atic lipase was 0.68,which was below 1.Conclusion Both vitexin and isovitexin had inhibitory effect on pancreatic lipase,and their inhibition types were reversible competitive inhibition.The inhibitory ability of their combined use was stronger than using alone,indicating that they had a good synergistic inhibitory effect on lipase.
