Protective effect and mechanism of Qizhi Yishen Capsules on HK-2 cell injury induced by high glucose

Abstract：Objective To study the protective effect and mechanism of Qizhi Yishen Capsules on HK-2 cell injury in-duced by high glucose.Methods HK-2 cells were randomly divided into control group(DMEM medium),model group(DMEM medium+60 mg·mL-1 glucose solution)and optimal dose group of Qizhi Yishen Capsules(DMEM medium+60 mg·mL-1 glucose solution+300 μg·mL-1 Qizhi Yishen Capsules lyophilized powder solution).The survival rate of HK-2 cells was detected by CCK-8 method,the content of ROS was detected by reactive oxygen species detection kit,the apoptosis and cell cycle were detected by flow cytometry,and the content of ATP was detected by ATP detection kit.ResuIts Compared with the control group,the cell survival rate of the model group was significantly inhibited,with significant difference(P<0.01).Compared with the model group,the cell survival rate of each Qizhi Yishen Capsules group was increased,and there was a significant difference(P<0.01).Compared with the control group,the apoptosis rate of HK-2 cells in the model group was significantly increased,the proportion of G0/G1 phase was increased(P<0.000 1),the proportion of G2/M phase+S phase was decreased(P<0.000 1),the content of ROS was significantly increased(P<0.000 1),and the content of ATP was signifi-cantly decreased(P<0.000 1).Compared with the model group,the proportion of apoptosis in the Qizhi Yishen Capsules group was decreased,the proportion of G0/G1 phase was decreased(P<0.05),the proportion of G2/M phase+S phase was increased(P<0.05),the ROS content in the cells was decreased(P<0.05),and the ATP content was increased(P<0.01).ConcIusion After the intervention of Qizhi Yishen Capsules,the inhibition of high glucose on the survival rate of HK-2 cells was re-duced,the proportion of G2/M phase+S phase was increased,the content of ROS was decreased,and the level of ATP was in-creased,indicating that Qizhi Yishen Capsules has a significant protective effect on HK-2 cell injury induced by high glucose.
