Abstract:Atherosclerosis is an early pathological change of many cardiovascular diseases.Lipid metabolism disorder is one of the most important risk factors of atherosclerotic cardiovascular disease(ASCVD).Proprotein convertase subtilisin/kexin type 9(PCSK9)is a key protein in lipid metabolism. It can bind to low-density lipoprotein receptor(LDLR)and degrade LDLR to prevent the clearance of low-density lipoprotein, resulting in increased low-density lipoprotein cholesterol(LDL-C)levels,and increasing risk of cardiovascular disease.PCSK9 is also involved in the production of inflammatory cytokines, endothelial dysfunction and the formation of atherosclerotic plaques.A number of studies have shown that PCSK9 is a promising new target for the treatment of ASCVD.PCSK9 inhibitors are currently a new type of drug for the treatment of hypercholesterolemia. This article reviewed the role of PCSK9 inhibitors in atherosclerosis and the research progress of drugs,so as to provide reference for clinical medication.
