Abstract:Objective To establish HPLC-DAD multi-wavelength switching fingerprint of Atractylodis Macrocephalae Rhizome(AMR)before and after stir-baking with soil(SAMR),and compare the differences in chemical fingerprint peaks. Methods The HPLC fingerprints of 17 batches of AMR and 17 batches of SAMR were determined, and the similarity anal-ysis,cluster analysis, and OPLS-DA analysis were used. Results In their HPLC fingerprints, nine common peaks were i-dentified in AMR and ten common peaks were identified in SAMR.Six components, neochlorogenic acid, chlorogenic acid, atractylenolide Ⅰ,Ⅱ,Ⅲ and atractylon were identified. The peak areas of peak 1,peak 4(atractylenolide Ⅲ),and peak 8(atractylenolide Ⅱ)increased after stir-baking with soil. The peak areas of peak 2(neochlorogenic acid),peak 3(chloro-genic acid),peak 5,peak 6,peak 7,peak 9(atractylenolide Ⅰ),and peak 10(atractylon)decreased after stir-baking with soil. Conclusion The established HPLC fingerprint method can provide an experimental basis for further standardizing the processing technology of SMAR,formulating the specific quality standard, and studying the processing principle of SAMR.
