The research on the effect and mechanism of Periplaneta americana extracts on reversing multidrug resistance of BEL-7402/5-Fu

Abstract：Objective To investigate the effect and mechanism of extracts from Periplaneta americana extract CⅡ-3 and skim cream on reversing multi-drug resistance in BEL-7402/5-Fu.Methods By measuring the cell growth curve and apoptosis rate of BEL-7402/5-Fu and the uptake of P-gp in BEL-7402/5-Fu cells,the reversal of BEL-7402/5-Fu by Periplaneta americana extract in vitro was investigate.Results With the concentration increase of CⅡ-3 and skim cream,the inhibition rate,reversal fold and apoptosis rate of BEL-7402/5-Fu increased gradually.There was no significant difference between CⅡ-3 and skim cream,the results showed that CⅡ-3 and skim cream on the reversal of drug-resistant cells and induction of apoptosis was equivalent.CⅡ-3 and skim cream can weaken the P-gp efflux function,and intracellular drug accumulation more skim cream stronger than CⅡ-3,intracellular drug accumulation of more.Conclusion The Periplaneta americana extract CⅡ-3 and skim cream could effectively reverse the multidrug resistance of BEL-7402/5-Fu cells,the reversal mechanism was mainly through the induction of drug-resistant cell apoptosis and inhibition of P-gp protein expression to achieve.
