Growth inhibition and apoptosis in human colorectal cancer cells induced by 5F - extracted from Pteris semipinnata L.

Abstract:Objective To examine the antiproliferative and proapoptotic effects of 5F(ent - 11α - hydroxy - 15 - oxo- kaur - 16 - en - 19 - oic - acid),a compound isolated from Pteris semipinnata L.(PsL),on HCT - 116 colorectal cancer (CRC)cells. Methods Cytotoxicity was examined using a MTT method. Cell apoptosis was determined through expression of representative apoptosis - related proteins and Annexin V/ PI staining. Reactive oxygen species(ROS)level was meas-ured using standard protocols. Results 5F inhibited the proliferation of HCT - 116 cells in a concentration - and time - de-pendent manner. 5F increased the ratio of Bax/ Bcl - 2 in HCT - 116 cells was identified by western blotting analysis. 5F could enhance both the inhibitory effect of cisplatin on growth of HCT - 116 cells and the level of apoptosis induced by cis-platin in HCT - 116 cells. 5F did not increase ROS,but reduced ROS production induced by cisplatin. Conclusion 5F can induce CRC cell apoptosis and therefore represents therapeutic potential for cancer treatment.
