The progress on pharmacological and clinical research of small molecule inhibitors regorafenib

Abstract:Pharmacology,pharmacokinetics,clinical study and safety of regorafenib in the treatment of colorectal cancer and gastrointestinal stromal tumors were reviewed in the paper. The main refeience to foreign - related registration and clini-cal data of regorafenib. Clinical trials showed the Overall Survival(OS)and Progression - Free - Survival(PFS)was sig-nificantly increased in patients treated with regorafenib. The most serious adverse drug reactions in patients receiving Stivar-ga were severe liver injury,haemorrhage and gastrointestinal perforation. The most frequently observed adverse drug reactions (≥30% )in patients receiving Stivarga were asthenia/ fatigue,decreased appetite and food intake,hand foot skin reaction, diarrhoea,weight loss,hypertension and dysphonia.
