Study on the pharmacokinetic of loganin in Liuwei Dihuang Pills in rats

Abstract：Objective To establish an HPLC - MS methods for the determination of loganin in rats plasma and to study the pharmacokinetic of loganin after administration of Liuwei Dihuang Pills. Methods 6 Wistar rats were administrated with Liuwei Dihuang Pills suspension(10 g·kg - 1 ). The blood samples were collected as scheduled time and analyzed by HPLC - MS. The pharmacokinetic parameters of loganin were calculated by DAS 2. 0 pharmacokinetic software. Results The main parameters of loganin in Liuwei Dihuang Pills were shown as follows:t1 / 2(4. 398 ± 1. 633)h,Tmax(2. 083 ± 1. 021)h,Cmax(369. 314 ± 53. 452)ng·mL - 1 ,AUC0 ~ 24(2 275. 174 ± 553. 825)ng·mL - 1 ·h,AUC0 ~ ∞(2 343 ± 601. 313)ng·mL - 1 ·h. Conclusion The HPLC - MS method was sensitive and selective for determination of loganin in plasma. A bimodal phenomenon was observed in the plasma prole after administration of Liuwei Dihuang Pills.
