Acute toxicity experimental study of mussaendoside U administered by caudal vein injection in mice

Abstract：Objective To study the acute toxicity reaction and determine the median lethal dose and its 99% confi-dence limit when mice were administered mussaendoside U by caudal vein injection. Methods Mice received administra-tion by caudal vein injection of mussaendoside U,pilot test was carried out to look for minimum 0% lethal dose and maxi-mum 100% lethal dose. And then,10 groups mice were injected mussaendoside U concentration according 1:0. 8 geometric dilution solution of 0. 01 mL · g-1 ,dose of each group was between minimum 0 lethal dose and maximum 100% lethal dose. The situations of death by poisoning in each group within 15 days after the treatment were observed. Results The death rates of lowest dose group and highest dose group were within 20% ~70% while administration dose was 128~760 mg·kg-1. There was approximate linear relationship between logarithm of dosage and death probability. Conclusion The median lethal dose of mice administered mussaendoside U by caudal vein injection was 286. 327 mg·kg-1 ,and its 99%confidence limit was(286. 327 ± 76. 189)mg·kg-1.
