Studies on the preparation and in vitro release characteristics of Flurbiprofen(poly lactic-co- glycolic acid)Microspheres for Intra-articular Injection

Abstract:objective To prepare Flurbiprofen( FP)Microspheres for Intra-articular Injection and study its release characteristics in vitro. Methods Poly( lactic-co-glycolic acid )( PLGA)was used as carrier material and the micro-spheres were prepared by emulsion-solvent evaporation method. The preparation process of FP microspheres was optimized by orthogonal design. In vitro release performance was got by using dialysis method in pH 7. 4 phosphate buffer,and FP was measured by UV spectrophotometry. Results The concentration of PLGA was a very significant factors affecting the entrap-ment efficiency. The particle sizes of FP microspheres were focused on 1. 5~24. 3μm,the mean size was 10. 6μm,and the drug loading was 7. 1%( W/W),entrapment efficiency was 92. 7%,respectively. The in vitro release characteristic of FP microspheres was in coincidence with Higuchi equation. Release time was significantly longer than raw material medicine. Conclusion The size distribution of FP microsphere was focalized,and it showed obvious sustained effect in vitro. FP mi-crospheres were suitable for intraarticular injection use.
