Determination of chlorogenic acid and galuteolin With different processing methods in flos lonicerae by HpLC

Abstract:Objective To investigate the effect of different processing methods on the content of chlorogenic acid and galuteolin in flos lonicerae and the quality of flos lonicerae. Methods The determination of chlorogenic acid was performed on Agilent Zorbax SB - C18 column(4. 6 mm × 250 mm,5 μm). The mobile phase was acetonitrile - 0. 4% phosphate(15:85)with the flow rate of 1. 0 mL·min - 1 . The detection wavelength was 327 nm and the column temperature was room temperature. The determination of galuteolin was performed on Hypersil Phenyl - 2 column(4. 6 mm × 150 mm,5 μm). The mobile phase was acetonitrile - 0. 5% glacial acetic acid with binary gradient elution,acetonitrile 10% ~ 20% within 0 ~ 15 min,acetonitrile 20% within 15 ~ 30 min,acetonitrile 20% ~ 30% within 30 ~ 40 min. The flow rate was 1. 0 mL·min - 1 , the detection wavelength was 350 nm and the column temperature was 40 ℃ . Results Different processing methods had certain effects on the content of chlorogenic acid and galuteolin in flos lonicerae. Conclusion It was suitable to use the pro-cessing of drying method and microwave in the production. And there was close relationship between the quality and appear-ance of color.
