Screening of rheumatoid arthritis biomarkers based on bioinformatics

Abstract:To assist in the diagnosis and treatment of rheumatoid arthritis(RA),we employed bioinformatics approaches to identi-fy and validate key biomarkers associated with RA.Firstly,the transcriptomic dataset of RA was obtained from the Gene Expression Omnibus(GEO)database.Differentially expressed genes were analyzed and screened,followed by functional enrichment analysis to in-vestigate their biological roles.Subsequently,six hub genes(CCR7,CD2,CD27,CD8A,GZMB and IL7R)were identified in the protein-protein interaction(PPI)network,and the expression levels of these genes were verified using independent datasets and real-time quantitative polymerase chain reaction(RT-qPCR).The study demonstrated that miR-9-5p and IL7R were the core components of a competing endogenous ribonucleic acid(CeRNA)network,and six key genes were identified.Among them,IL7R and miR-9-5p might play a crucial role in the progression of RA.This research can provide a new direction for the diagnosis and treatment of RA.
