The Relationship of Insulin and PI-3 K/AKT Pathway after Mouse′s Spontaneous Subarachnoid Hemorrhage

Abstract:To investigate whether insulin plays a role in the anti -apoptotic in mouse spontaneous subarachnoid hemorrhage (SAH),if so,which signaling steps are involved in this process.SAH was induced in male Wistar mouses by puncture of internal ca-rotid artety.Seventy male Wistar mouse with 28~32 g were randomly divided into 4 groups:Sham (control)group,SAH group,Ins (SAH+insulin)group,PD (SAH+insulin+PD98059)group.(1)Neurological examination:mouse′s psychosis,feeling and sports function were observed and recorded.(2)The morphological changes of the neurons were studied by light microscope use HE method. (3)RT PCR were preformed for detecting the expression change of p-AKT and p-ERK m RNA.(4)The expression of p-AKT and p-ERK were observed in the mouse brain with western blot method.The results showed:(1 )Neurons in the hippocampal CA1 re-gion after SHA showed typical apoptotic features under light microscope,but Ins group was better than that of SHA group,while PD group was no better than that of SAH group.(2)Both of the expression of p-AKT and p-ERK mRNA were higher than that of Sham group,but in Ins group,both of them were higher that of SAH group,while PD group them were not higher than that of SAH group. (3)Western blot result showed that the protein expression level of p-AKT,p-ERK was as same as the expression level of mRNA.In-sulin can block apoptosis by increase phosphatidylinositol 3-kinase (PI-3K)in spontaneous subarachnoid hemorrhage and p-ERK may play an important role in this process.
