Abstract:To overcome the bottleneck in asexual propagation of Liriodendron tulipifera,this study used apical and axillary buds from young shoots as explants for tissue culture.A cyclic disinfection method involving"sub culturing and disinfection procedures",was established,This method effectively addressed common challenges during the culture process,such microbial contamination,browning,leaf abscission,and excessive callus formation during bud elongation.The optimal media formulations were determined as follows:Shoot proliferation medium:1/2 MS+0.5 mg/L KT+0.1 mg·L-1 6-BA+0.1 mg·L-1 NAA.Bud elongation medium:WPM+0.1 mg·L-1 6-BA+1.0 mg·L-1 IBA+100 mg·L-1 VC.Rooting medium:WPM+0.02 mg·L-1 6-BA+1.5 mg·L-1 IBA.After 40 days of culture on the rooting medium,each shoot produced 1-4 roots,achieving a rooting rate of 73.3%.
