Tissue Culture and Plantlet Regeneration of Tall Fescue(Festuca aruudiuacea)

Abstract:In order to establish an efficient plant regeneration system,MS was used as the basic medium appended with different concentrations of 2,4-D,the effects of 2,4-D on tissue culture and culture responses were investigated using the seed,stem base,immature inflorescences and leaf tips as explants of tall fescue(Festuca aruudiuacea).Meanwhile,the callus propagation,differentiation culture medium and the effects of different medium types,NAA concentration and light conditions on rooting of tall fescue were studied.The results indicated base was the best organ for callus induction,with the highest callus induction rate of 44.3%,the optimal concentration of 2,4-D was 4.0mg/L,callus performed best.Subc-ulture medium of"MS+2,4-D4.0mg/L"after twice multiplication the callus had the highest multiplication coefficient(16.13)and the lowest callus mortality."MS++6-BA3.0mg/L+NA-A0.5mg/L"had the obvious differentiation-inducting effect on callus and resulted the high-est regeneration rate(78.1%).In the stage of rooting stage,MS had a better efficiency than 1/2MS and the best culture medium was MS+0.3mg/LNAA,dark culture conditions weremore conducive to rooting induction of tissue culture seedlings than light culture.This study established the efficient regeneration system of F.arundinacea.
