Research on Tissue culture technique of Hemerocallis hybrida ‘Big Eye’

Abstract：The young shoots of Hemerocallis hybrida ‘Big Eye’were used as explants in the study.Through the plant tisG sue culture techniques,the experiment studied the establishment of H .hybrida ‘Big Eye’rapid propagation system.Using MS as the basic medium supplemented with different concentrations of 6-BA and NAA plant growth regulating substances,in orG der to study the effect of different culture conditions on H .hybrida ‘Big Eye’in vitro starting culture,proliferation and rooG ting culture.The results showed that the optimum initiation and multiplication medium was MS+6-BA2.0mg/L+NAA0. 5mg/L,adventitious bud differentiation rate was 76.7%,and adventitious bud average growth was 2.90cm after 40d;the best multiplication culture medium was MS+6-BA4.0mg/L+NAA1.0mg/L,and proliferation bud number in 4.97 on average;the best rooting medium was 1/2MS+IBA0.25mg/L ,the rate of survival was 100%.
