Apoptosis and Lipolysis of White Adipocytes Induced by Neuropeptide Y-Y5 Receptor Antisense Oligodeoxynucleotides
Abstract:Objective:To investigate the influence of central administration of neuropeptideY-Y5 receptor antisense oligodeoxynucleotides (ODNs) on body weight, fat pads of SD rats, and the ef-fects of white adipocytes lipolysis and apoptosis. Methods:Y 5 receptor antisense, sense, mismatcbedODNs or vehicle was intracerebroventricularly ( i. c. v. ) injected. Average adipocyte area was calculat-ed. DNA ladders were measured to evaluate adipocyte apoptosis, and RT-PCR was used to analyse theexpresswn of Bcl-2 and Box gene. Results:Central administration of Y5 antisense ODNs significant-ly decreased body weight, and average adipocyte area. DNA fragmentation was present after electrophore-sis at epididymal adipose tissue. The expression of Bcl-2 gene was downregulated, while the expressionof Box upregulated. Conclusion:Lipolysis and adipocyte apoptosis may be important mechanisms forY5 antisense therapy.
