Study on the mechanism of sub-anesthetic dose of sevoflurane attenuate yeast poly-saccharide-induced RAW 264.7 macrophage injury

Abstract:Objective To investigate the effects of sub-anesthetic doses of sevoflurane on the proliferation,apoptosis and oxidative stress levels of yeast polysaccharide(zymosan)-induced RAW264.7 macrophages.Methods Mouse RAW264.7 macrophages were trea-ted with various concentrations of zymosan(0,2.5,5,10,20,40,80,160 μg/mL)for 24 h.The CCK-8 kit was used to assess cell viability and determine the half maximal inhibitory concentration(IC50)of zymosan on macrophages.RAW264.7 macrophages were ran-domly divided into three groups,a blank control group(Control group),a zymosan-treated group(Zym group),and a zymosan+sub-anesthetic dose of sevoflurane-treated group(Zym+Sevo group).The Edu cell proliferation assay kit,mitochondrial membrane potential detection kit(JC-1),and reactive oxygen species(ROS)detection kit were employed to evaluate cell proliferation,apoptosis and oxida-tive stress levels,respectively.Results The CCK-8 assay results showed that the IC50 of zymosan on macrophages was 31.80 μg/mL.Zymosan stimulation led to a decrease in the EdU-positive cell rate,a reduction in mitochondrial membrane potential,and an increase in ROS levels(P<0.01).After treatment with a subanesthetic dose of sevoflurane,compared to the Zym group,the Zym+Sevo group exhibi-ted an increase in the EdU-positive cell rate,an elevation in mitochondrial membrane potential,and a decrease in ROS levels(P<0.01).Conclusion Sub-anesthetic doses of sevoflurane can attenuate the adverse effects of zymosan-induced inhibition of macrophages prolifera-tion,increased apoptosis and elevated oxidative stress levels.
