Abstract:Objective To investigate the relationship between cytotoxic T lymphocyte-associated antigen-4(CTLA-4)gene polymorphism and breast cancer in female patients with type 2 diabetes mellitus(T2DM).Methods A total of 337 female T2DM patients admitted from July 2022 to March 2025 were selected as the study subjects, and they all underwent CTLA-4 gene polymorphism testing. They were divided into breast cancer group and non-breast cancer group according to the prevalence of breast cancer in female T2DM patients. The clinical data and CTLA-4 gene polymorphism of the two groups were compared, and the relationship between CTLA-4 gene polymorphism and breast cancer in women with T2DM was analyzed. Results Among 337 female T2DM patients,42 patients had breast cancer and the prevalence rate was 12.46%.The glycosylated hemoglobin(HbAlc),homeostasis model assessment of insulin resistance(HOMA-IR),distribution of the AA genotype at the CTLA-4 gene-49 locus and A allele frequencies in breast cancer group at admission were higher than those in non-breast cancer group(P<0.05,P<0.01).Multivariate logistic regression analysis showed that HbAlc,HOMA-IR and the AA genotype and A allele of CTLA-4 gene-49 locus were risk factors for breast cancer in women with T2DM(P<0.01).Conclusion The AA genotype and A allele of CTLA-4 gene-49 locus are associated with breast cancer in women with T2DM.
