The expression of microRNA-27a and microRNA-21 in peripheral blood of patients with myasthenia gravis and their correlation with AChR-Ab and IL-6

Abstract:To investigate the expression and significance of microRNA-27 a and microRNA-21 in peripheral blood serum of patients with myasthenia gravis, total of 108 patients diagnosed as myasthenia gravis (MG) for the first time in our hospital from September 2015 to March 2018 were selected as the MG group, and 73 healthy people were selected as the normal group. The relative expression of microRNA-27 a and microRNA-21 in peripheral blood serum of MG and normal groups were detected by RT-PCR technology, while the expression level of AChR-Ab and IL-6 were detected by ELISA technology. Data showed that there was no significant difference in baseline databetween MG group and normal group (P>0.05). The relative expression of microRNA-27 a in peripheral blood of patients in MG group (0.89±0.25) was lower than that in normal group (2.16±0.34), while the relative expression of microRNA-21 in MG group (3.78±0.57) was higher than that in normal group (1.05±0.28) (P<0.01); the expression levels of AChR-Ab and IL-6 in MG group were (1.12±0.45) nmol/L and (180.88±40.80) ng/ml, respectively, which were higher than those (0.28 nmol/L±0.12 nmol/L and 115.46 nmol/L±31.27 ng/ml) in normal group (P<0.01). Correlation analysis showed that microRNA-27 a was negatively correlated with AChR-Ab and IL-6 (r=-0.823,-0.716, P<0.01), while microRNA-21 was positively correlated with AChR-Ab and IL-6 (r=740, 0.556, P<0.01). In conclusion, there are differential expression profiles of microRNA-27 a and microRNA-21 in peripheral blood serum of patients with myasthenia gravis, and the two were are correlated with AChR-Ab and IL-6, but the specific mechanism remains to be confirmed by further studies.
