Optimization of response surface method for polysaccharides extraction from Radix Pseudostellariae residues

Abstract:To improve the utilization rate of Radix Pseudostellariae, we optimized techniques of extracting polysaccharides from Radix Pseudostellariae dregs. The Radix Pseudostellariae residues extracted saponin was used as material to extract polysaccharides. Based on monofactorial experiments, main factors were selected from the Plackett-Burman experiments according to the yield of polysaccharides, and further optimized by a Box-Behnken response surface test. The optimized extraction conditions included material to liquid (30% ethyl alcohol) at 1:30 (g/ml), 80 ℃ for 115 min. Under the conditions, the yield and purity of polysaccharides were 6.01%±0.03% and20.80% ±0.13%, respectively. Compared to the direct extraction polysaccharides from Radix Pseudostellariae with a yield of 6.52%±0.06% and a purity of 17.56%±0.09%, the polysaccharides yield of residues was similar, while the purity was improved. In conclusion, the developed extraction technique can effectively improve the utilization level of Radix Pseudostellariae, and has good application prospect.
