Study on the anti-inflammatory effect and mechanism of Salvia plebeia R.Br. ethyl acetate extract on LPS-induced RAW264.7 cell

Abstract:To investigate the anti-inflammation effect and mechanism of Salvia plebeia R. Br. ethyl acetate extract (LZC) in vitro. Inflammatory factor release of PGE2, NO, TNF-α and IL-6 and the protein and m RNA expression of TLR4, MyD88 and NF-κB in LPS-induced RAW264.7 cell treated with LZC were measured with enzyme linked immunosorbent assay, qPCR and western blotting, respectively. MTT method were used to assay the effect of LZC on the viability of RAW264.7 cell. 6.25-25 μg/ml of LZC could significantly inhibit PGE2, NO, TNF-αand IL-6 levels in the LPS-induced RAW264.7 cell. Moreover, 6.25-25 μg/ml of LZC could obviously downregulate the protein and mRNA expression of TLR4, MyD88 and NF-κB in LPS-induced RAW264.7 cell. Saluia plebeia R.Br. ethyl acetate extract could inhibit the inflammatory reaction of RAW264.7 cells induced by LPS via TLR4 signal pathway way.
