Th22 cells promote ovarian cancer progression by releasing IL-22 to activate the STAT3 pathway

Abstract:To evaluate the roles of IL-22 in ovarian tumor, we detected the expression of IL-22 in tumor tissues and adjacent tissues of different stages. The frequencies of CD4+cells and Th22 in tumor tissue, patient peripheral blood and normal human peripheral blood were detected by flow cytometry; the localization of IL-22 in cells was detected by flow immunofluorescence laser confocal microscopy. Western blot assay was used to detect the expression of STAT3 and phosphorylated STAT3 (p-STAT3) in ovarian cancer tissues and paracancer tissues; cell co-culture assay was used to evaluate whether IL-22 activated STAT3 pathway and promoted ovarian cancer development. Data revealed that the expression of IL-22 in ovarian cancer tissues were higher than that in adjacent tissues. The frequencies of CD4+and TILs-Th22 were increased significantly in the tumor tissue and positively correlated with the tumor stage. IL-22 in ovarian cancer tissue was secreted by CD4+cells and cell co-culture showed that IL-22 activated STAT3 pathway and induced downstream molecular expression. Therefore, Th22 cells can release IL-22 and activate STAT3 signaling pathway to promote ovarian cancer development. The results from this study provide a new potential strategy for ovarian cancer treatment.
