Ebastine inhibites the degranulation and cytokine release of mast cells in vitro

Abstract:This study was performed to investigate the effect of ebastine on the degranulation and cytokine release of mast cell induced by mast cell activator C48/80 in vitro.MTT assay was used to detect the proliferation of mast cells,while the effect of ebastine on the granulation of mast cells was detected by β-hexosaminidase degranulation kit.The secretion of TNF-α,IL-4,IL-1β,LTB4,and histamine were detected by ELISA and the mRNA expression of eotaxin,TNF-α,IL-4,IL-1β,IL-5 and IL-6 were detected by reverse transcriptionquantitative PCR.Data showed that the doubly increased doses of ebastine remarkably inhibited degranulation,the indicated cytokines and chemokines secretion and mRNA expression from mast cells,among of which the four-folded dose of ebastin demonstrated the most significant inhibition effects on degranulation.Therefore,results in this study indicate that the doubly increased dose of ebastin can inhibit the degranulation and cytokine production of mast cells,and the four-fold dose of ebastin shows the most significance of inhibition,suggesting the potential clinical application of this dose of ebastin for urticaria treatment.
