Neutrophils act as tumor-promoting cells by secreting matrix metalloproteinase 2 and 9

Abstract:Tumor-associated neutrophils (TANs) consist of N1 and N2 types.N1 TANs inhibit the development of tumor,while N2 TANs facilitate the development of tumor.However,the mechanism of N2 type is unclear.This study was performed to investigate the effects of gastric cancer microenvironment or interferon β inhibition on matrix metalloproteinase 9 (MMP-9) and MMP-2 secretion from normal human neutrophils.Peripheral blood neutrophils of 51 normal donors were separated,and incubated with IFN-β antibody of 5 μg/ml (antibody group) or 50% gastric cancer cell MGC-803 culture medium (supernatant group).The mRNA and protein levels of MMP-9 and 2 generated from neutrophils were detected using real-time PCR and ELISA.And enzymatic activity of MMP-2 and MMP-9 were analyzed by gelatin zymography assay.Data showed that abundance of MMP-2 and MMP-9 mRNA in antibody group and supernatant group were more than that in control group (P<0.01);the concentration of MMP-2 and MMP-9 protein in antibody group and supernatant group were higher than that in control group (P<O.01).Enzyme activity of MMP-2 and MMP-9 in antibody group and supernatant group were also stronger than that in control group (P<0.01).Our study suggests that gastric cancer microenvironment and interferon β inhibition can induce neutrophils secreting MMP-9 and MMP-2 to promote tumor development,acting as N2 type TANs.
