miR-143-3p suppresses IL-6-induced angiogenesis of HLMVEC cells via targeting IL-6R/STAT3 signaling

Abstract:This study aimed to explore the role of miR-143-3p in IL-6-induced angiogenesis of HLMVEC cells and the mechanism.Firstly,100 ng/ml of recombinant IL-6 protein was used to treat HLMVEC cells.Tube formation assay was used to measure the effect of IL-6 on the number of tubes formed;the effects of IL-6 on miR-143-3p and VEGF mRNA expression were analyzed by Real-time PCR.VEGF protein levels were determined by Western blotting.Then,miR-143-3p was overexpressed by miR-143-3p mimics,and the effects of miR-143-3p on tube formation,VEGF mRNA and protein levels were assayed.Dual luciferase reporter gene assay was used to determine the relationship between miR-143-3p and IL-6R;the effect of miR-143-3p overexpression on IL-6R mRNA expression was measured by Real-time PCR.Western blot was used to detect the protein levels of IL-6R,STAT3,and phosphorylated STAT3 (p-STAT3).Data showed that IL-6 could increase the number of tubes formed and VEGF expression,but reduce miR-143-3p level,miR-143-3p overexpression inhibited IL-6-induced tube formation and the expression of VEGF,IL-6R,and p-STAT3,but did not change STAT3 levels.Additionally,IL-6R is a target gene of miR-143-3p.In conclusion,miR-143-3p could inhibit IL-6-induced angiogenesis of HLMVEC cells,which may function via suppressing IL-6R/STAT3 signaling.
