Abstract:This study was performed to investigate the effect of different treatment regimens on the level of myeloid-derived suppressor cells (MDSCs) subsets in peripheral blood mononuclear cells (PBMCs) of patients with digestive tract cancers.Forty patients and 10 healthy people were recruited from the Second Affiliated Hospital of Zhengzhou University.Peripheral blood samples were collected before and after chemotherapy, surgery or surgery combined with chemotherapy.Granulocytic MDSCs (G-MDSCs) and monocytic MDSCs (M-MDSCs) levels were detected by flow cytometry;while Arg-1 and iNOS released by MDSCs were detected by ELISA.Data showed that the levels of G-MDSCs and M-MDSCs in peripheral blood of patients were significantly higher than those in healthy people, and decreased after treatment;the level of iNOS released by MDSCs was decreased after surgery or chemotherapy.Moreover,G-MDSCs level was positively related to lymph node metastasis and tumor size (≥5 cm),while M-MDSCs level was negatively related to cancer differentiation. In conclusion,MDSCs subsets in peripheral blood of digestive tract tumor patients are increased and can be reduced by chemotherapy and/or surgery.Levels of iNOS of MDSCs are decreased after surgery or chemotherapy.G-MDSCs are associated with tumor size and lymph node metastasis, while M-MDSCs are related to the degree of tumor differentiation.
