Inhibitory effects of baicalin on NLRP3 inflammasome activation and pyroptosis and the underlying mechanism

Abstract:This study was performed to explore the influences of baicalin on ATP-induced inflammasome activation and pyroptosis in LPS-primed murine macrophages (J774A.1 or peritoneal macrophages) and the underlying mechanism.Peritoneal macrophages were elicited by intraperitoneal injection of 30 g· L-1 thioglycollate medium into C57BL/6 mice;ATP was used to activate the NLRP3 inflammasome in LPS-primed macrophages.The protein levels of IL-1 β,caspase-1 and HMGB 1 expressed in cell lysates and culture supernatants were evaluated by Western blotting;soluble IL-1β in the culture supernatants was determined by bead-based immunoassay (CBA).Influences of baicalin on pyroptosis induced by LPS plus ATP were analyzed by propidium iodide (PI) staining.Data showed that baicalin dose-dependently inhibited caspase-1 activation,mature IL-1β and HMGB1 secretion in the LPS-primed macrophages upon ATP stimulation.Baicalin also suppressed pyroptosis in murine macrophages stimulated with LPS plus ATP.Adenyl cyclase (AC) inhibitor MDL12330A or protein kinase A (PKA) inhibitor H89 could significantly reverse baicalin-mediated suppression on ATP-induced pyroptosis.In conclusion,baicalin suppresses NLRP3 in flammasome activation and pyroptosis,probably through modulating PKA activity in macrophages.
