The expression and function of soluble B7-H3 in healing process of human traumatic brain injury combined with fracture

Abstract:To investigate the expression of soluble B7-H3 (sB7-H3) in the peripheral blood of patients with traumatic brain injury (TBI) combined with fracture,fracture only,or TBI only,and study the effects of sB7-H3 on differentiation of mesenchymal stem cells (MSCs) to osteoblast,the expression of sB7-H3 and fracture healing marker Cbfa1 in peripheral blood of patients with TBI combined with fracture,fracture only,TBI and normal volunteers were detected by ELISA,and the effect of recombinant B7-H3 protein on osteogenic differentiation of MSCs were examined by Alizarin red and real-time PCR.Data showed that the expression of sB7-H3 in patients with TBI combined with fracture or TBI were significantly higher than that in fracture only and normal volunteers.Moreover,the concentration of sB7-H3 in peripheral blood was correlated with the severity of traumatic brain injury.And recombinant B7-H3 protein could promote the differentiation of MSCs into osteoblasts in a dose dependent manner.In conclusion,the elevated sB7-H3 in peripheral blood caused by TBI promotes fracture healing.
